چکیده

Introduction Osteoarthritis (OA) is a degenerative joint disease characterized by articular cartilage degradation, subchondral bone sclerosis and osteophyte formation. Recent studies have demonstrated that an increase in subchondral bone turnover is accompanied by specific architectural changes in the trabecular bone in OA joints. Furthermore, epidemiological studies have demonstrated positive correlation between subchondral bone sclerosis and OA progression. These observations suggested a role for subchondral bone changes in the initiation and/or progression of OA, raising the possibility that early intervention that reduces subchondral bone remodeling might retard the progressive loss of articular cartilage. To investigate the potential contribution of bone turnover to OA progression, we evaluated the effect of Alendronate (ALN), a potent bone resorption inhibitor, on cartilage degradation and periarticular bone changes in the rat anterior cruciate ligament transection (ACLT) model of OA.

Effect of alendronate on post-traumatic osteoarthritis induced by anterior cruciate ligament rupture in mice

INTRODUCTION Previous studies in animal models of osteoarthritis suggest that alendronate (ALN) has antiresorptive and chondroprotective effects, and can reduce osteophyte formation. However, these studies used non-physiologic injury methods, and did not investigate early time points during which bone is rapidly remodeled prior to cartilage degeneration. The current study utilized a non-invasiv...

Molecular changes in articular cartilage and subchondral bone in the rat anterior cruciate ligament transection and meniscectomized models of osteoarthritis

BACKGROUND Osteoarthritis (OA) is a debilitating, progressive joint disease. METHODS Similar to the disease progression in humans, sequential events of early cartilage degradation, subchondral osteopenia followed by sclerosis, and late osteophyte formation were demonstrated in the anterior cruciate ligament transection (ACLT) or ACLT with partial medial meniscectomy (ACLT + MMx) rat OA models...

A Multiscale Model of the Cartilage Bone Interface to Investigate the Initiation of Osteoarthritis

The initiation of cartilage degeneration has been linked to the onset of subchondral bone remodelling and osteophyte formation at the cartilage-bone interface. As such, Osteoarthritis (OA) is defined as the thinning of cartilage and/or osteophyte formation. Cartilage adjacent to healthy bone has a characteristic stress distribution when loaded within a healthy range. This loading is then transf...

Bone Alkaline Phosphatase in Synovial Fluid Is Correlated to Radiographic Osteophyticchanges in a Canine Cranial Cruciate Transection Model of Osteoarthritis

INTRODUCTION: Osteoarthritis (OA) is considered to be a complex disease process that involves all components of the joint environment: cartilage, synovium, and subchondral bone. When there is joint injury, such as cranial cruciate ligament (CCL) rupture in dogs or humans, there are abnormal biomechanical stresses placed on the entire joint. Invariably, this leads to loss of the articular cartil...
